at the same time as checking.

3. diluted Aqueous extract is used (at a concentration of 100 ml/l) with the addition of

the same nutrients and at the same concentration as in control, according to the

the appropriate technical standards. In the case of tests with luminiscenčními

the bacteria Vibrio fischeri that means to 0.5 ml of the diluted sample

The test is performed with the seeds of lettuce Lactuca ' heads

sativa var. capitata, Sapphire. For the test are selected undamaged seeds

the same size, chemically unhandled. Seeds of lettuce can be

předklíčit in test container, on a layer of filter paper element

water for 24 h to 48 h at room temperature, without regulation

lighting. For the test are collected on sprouted seeds, respectively. with the root,

that is less than 2 mm.

To test the containers weigh 200 g to 300 g of the element under test

the sample of the waste (diluted in mass ratio of 1:9 artificial soil) or

control (artificial soil). The height of the layers of the sample in the container must be at least

3 cm. is laid out right-angle network, for example. 5 x 3 points. The created holes

approx. 0.5 cm to 1 cm deep with tweezers to evenly distribute after 15

germinated seeds of lettuce, root down. The seeds to a sample

the pattern is free, and thus prepared the container closed

the lid is placed in the thermostat with a temperature of 24 ° c + 40 ° c without access

light.

The test shall be carried out in three parallel series. After 120 h +-2 h

incubation salad gently separated from the sample and measure the and records

the length of all the roots in the test sample and checking with an accuracy of 1

mm.

The basis for the evaluation of growth inhibition test is the average length of the root

identified in the inspection and test sample. If the předklíčené seed

not created by the Constitution, does the this value to the mean as

zero. Coefficient of variation of parallel determinations may not exceed 20%.

The average length of the root of lettuce in the control must be at least 15 mm.

It is recommended to regularly perform the test with the reference substance.

Boric acid EC50 is determined using artificial soil, taking

the recommended value is in the range of the EC50 300.0 mg. kgSUŠ-1 to 650.0

mg. kgSUŠ-1.

To demonstrate the consistency of the laboratory test conditions, are to

each test included three root growth inhibition test containers

filled with sand, after 6 předklíčeného lettuce seeds.

The evaluation of the tests shall be carried out in accordance with the standard ČSN EN ISO 11269-1.

The recommended length of the root of the mean is 30 mm.
